- Will Q Biotech have more application specific cartridges available?
- Yes! We are currently working on IMAC cartridge for His-tag protein purification and a GST cartridge. Availability Q1 2025
- I am purifying recombinant proteins from E. coli. What kind of columns can be used with the open cartridge?
- Any column (pre-packed, such as HiTrap, or self-packed) that can be attached to the system using luer-lock connectors and can be run at low pressure (< 15 psi) can be used. Columns without luer-lock connectors can be used with an adaptor. For recombinant proteins from E. coli, common approach is to use His-tagged proteins. In this case, HisTrap or Talon columns from Cytiva, HisPure column from ThermoFisher work well. Pre-packed columns for other affinity tags are also available from these vendors.
- Can I use low pressure columns from Cytiva and BioRad combined with the open cartridge?
- Yes! The columns from Cytiva, BioRad, ThermoFisher and others can be easily used with the open cartridge. The columns often contain luer-lock adaptors for easy connection.
- Do you have any information of the actual application cases other than IgG purification?
- Our lab is currently working on several application notes, watch this space!
- Are any columns compatible with the open cartridge as long as they meet with specification of the Inceptum system (Flow rate: <50 mL/min, Max pressure: 15 psi) and can be connected with the system using standard Luer-lock?
- Correct! Any column designed for low pressure chromatography in a water- based buffer is compatible with the Inceptum system. Luer-lock adaptors are widely available from many vendors.
- Are there any precautions when selecting columns for use with the open cartridges?
- There are no precautions specific to Inceptum. Typical recommendations for column selection include:
- Pre-packed columns – make sure the thread to luer-lock adaptor has correct thread size.
- Avoid organic solvents that can damage polycarbonate as the UV and conductivity flow cell contains polycarbonate.
