During a recent demonstration of our Inceptum™ system at a major biopharmaceutical company, we used our Antibody purification cartridge to showcase the capabilities of the Next Generation Chromatography™. The result speaks for itself! Purification was as designed, less than 30 min to complete, from cartridge insertion to purified IgG. This includes 11 min for cartridge self-calibration, initialization and rinsing columns, 15 min to load sample onto protein G column and wash unbound proteins, and 3 min to elute IgG and cleanup by buffer exchange. The affinity purification and buffer exchange of antibody, resulting in >95% purity and available in ready to use format in less than 30 min is an industry-leading performance. For comparison, examples by market leading companies show 35-40 min for the affinity step alone, without the buffer exchange! (They never show how long it takes to prepare the system for the run! With The Next Generation Chromatography™ there is no setup or cleanup time).
Independent analysis of the purified IgG by customer shows equally outstanding performance: SEC analysis revealed less than 5% dimers and good purity by non-reducing and reducing SDS-PAGE.
